Researchers at the Wyss Institute for Biologically Inspired Engineering at Harvard University have developed a novel biomimetic strategy that delivers life-saving nanotherapeutics directly to obstructed blood vessels, dissolving blood clots before they cause serious damage or even death. This new approach enables thrombus dissolution while using only a fraction of the drug dose normally required, thereby minimizing bleeding side effects that currently limit widespread use of clot-busting drugs.

The research findings, which were published online today in the journal Science, have significant implications for treating major causes of death, such as heart attack, stroke, and pulmonary embolism, that are caused by acute vascular blockage by blood thrombi.

The inspiration for the targeted vascular nanotherapeutic approach came from the way in which normal blood platelets rapidly adhere to the lining of narrowed vessels, contributing to the development of atherosclerotic plaques. When vessels narrow, high shear stresses provide a physical cue for circulating platelets to stick to the vessel wall selectively in these regions. The vascular nanotherapeutic is similarly about the size of a platelet, but it is an aggregate of biodegradable nanoparticles that have been coated with the clot-busting drug, tissue plasminogen activator (tPA). Much like when a wet ball of sand breaks up into individual grains when it is sheared between two hands, the aggregates selectively dissociate and release tPA-coated nanoparticles that bind to clots and degrade them when they sense high shear stress in regions of vascular narrowing, such as caused by blood clot formation.

Disruption of normal blood flow to the heart, lung, and brain due to thrombosis is one of the leading causes of death and long-term adult disability in the developing world. Today, patients with pulmonary embolism, strokes, heart attacks, and other types of acute thrombosis leading to near-complete vascular occlusion, are most frequently treated in an acute care hospital setting using systemic dosages of powerful clot-dissolving drugs. Because these drugs can cause severe and often fatal bleeding as they circulate freely throughout the body, the size of the dosage given to any patient is limited because efficacy must be balanced against risk.

The new shear-activated nanotherapeutic has the potential to overcome these efficacy limitations. By targeting and concentrating drug at the precise site of the blood vessel obstruction, the Wyss team has been able to achieve improved survival in mice with occluded lung vessels with less than 1/50th of the normal therapeutic dose, which should translate into fewer side effects and greater safety. This raises the possibility that, in the future, an emergency technician might be able immediately administer this nanotherapeutic to anyone suspected of having a life-threatening blood clot in a vital organ before the patient even reached the hospital.

The inter-disciplinary and inter-institutional collaborative research team, which was led by Wyss Founding Director Donald Ingber M.D., Ph.D., and Wyss Technology Development Fellow Netanel Korin, Ph.D., also included Wyss postdoctoral Fellow Mathumai Kanapathipillai, Ph.D., as well as Benjamin D. Matthews, Marilena Crescente, Alexander Brill, Tadanori Mammoto, Kaustabh Ghosh, Samuel Jurek, Sidi A. Bencherif, Deen Bhatta, Ahmet U. Coskun, Charles L. Feldman, and Denisa D. Wagner from Brigham and Women's Hospital, Children's Hospital Boston, Harvard Medical School, the Harvard School of Engineering and Applied Sciences, and Northeastern University. Ingber is also the Judah Folkman Professor of Vascular Biology at Harvard Medical School and Children's Hospital Boston, and Professor of Bioengineering at Harvard's School of Engineering and Applied Sciences.

Commenting on the work, Ingber noted that "the vascular nanotherapeutic we developed that selectively becomes activated in regions of high shear stress, much like living platelets do, is a wonderful example of how we at the Wyss Institute take inspiration from biology, and how biomimetic strategies can lead to new and unexpected solutions to age-old problems that existing technologies can't address."

NBA Draft Grades: Perry Jones Earns High Marks For Oklahoma City Thunder

Better late than never is how the saying goes, so while the NBA Draft grades coming from Dennis Velasco at the Basketball Jones arrive after everyone else has filed their columns, it?s still a great read for basketball fans. The good news for the Oklahoma City Thunder is that Velasco loves their draft and the specific pick of Perry Jones.

Instead of just offering the quick take that everyone else does, Velasco goes the extra mile and gives depth on his opinion. That?s what makes this column a great read on every team. Specifically he loves the value of the pick and it becomes once again a potential impact addition by Sam Presti. Velasco writes:

This could end up being a great pick for the Thunder. During the beginning of Jones? freshman year, he was actually projected to be the first overall selection in many mocks. He has that much talent. However, there was a perceived dip in skill this season for Jones, who actually played out of position most of the season. It?s difficult to project what Jones will do with the Thunder as it will be dependent on where they play him.

Since Kevin Durant will man the three, you?d have to assume that Jones will play the four, which could mean the eventuality of Serge Ibaka being let go when he becomes a free agent after next season. That would be a mistake, but Jones is so talented that it would be a mistake not to start him. Time will tell, but this is easily a low risk-high reward pick at the end of the first round. It?s easy to love this pick, value-wise.

Pakistan: US drone strike kills 8 militants

(AP) ? U.S. missiles fired from a drone in a Pakistani tribal region near the Afghan border killed eight suspected militants early Sunday, officials said, as the controversial American strikes continue despite Islamabad's persistent demands that they stop.

The latest attack killed fighters loyal to militant commander Hafiz Gul Bahadur, the officials said. Bahadur is believed by residents of the region to have an informal working relationship with the Pakistani army, refraining from targeting the security forces while focusing on U.S. and NATO forces in nearby Afghanistan.

The continued strikes, despite the likely political fallout, show Washington's confidence in the effectiveness of the drone program against al-Qaida and Taliban fighters who allegedly use Pakistan as a base.

Two Pakistani intelligence officials said four Hellfire missiles were fired at a house used by suspected militants in Dre Nishter village of North Waziristan. All the Pakistani officials spoke on condition of anonymity because they were not authorized to speak to the media.

The officials said some foreign militants belong to the Turkmenistan Islamic Movement were believed to have been killed, along with other local fighters from the Bahadur group. Militants from several central Asian countries have joined Afghans, Arabs, and others in Pakistan.

North Waziristan is one of several tribal areas along the border with Afghanistan that are hubs of militant activity. Bahadur controls most of North Waziristan.

The U.S. rarely talks publicly about the covert CIA-run drone program in Pakistan.

The drone attacks are a source of deep frustration and tension between the U.S. and Pakistan. Islamabad says they violate its sovereignty and also cause civilian casualties.

They complicate efforts to normalize the relationship between Washington and Islamabad, including reopening supply routes through Pakistani territory to NATO and American forces in Afghanistan. Islamabad blocked the routes after American airstrikes accidentally killed 24 Pakistani soldiers in November.

The U.S. in turn criticizes Pakistan for failing to crack down on fighters who stage attacks in Afghanistan.

Can long-distance relationships survive? | The Hand

Long-distance relationships: Can they work? It depends

Q: I know you usually advise readers not to sit around and wait for a significant other ? long-distance relationships (stink). I get that. Does the advice change if the significant other is headed to Japan for a year (so, a definite endpoint) and really seems dedicated to making it work, with regular flights back to the States, waking up early to phone/Skype, etc.?

I really like this guy ? we?ve been dating for two years, and have grown very close ? but I also don?t know if I?m taking on too much if I choose to wait for him, and I get that waiting around isn?t usually a good idea. Am I giving up a great thing by giving up on this, or am I saving myself pain in the long run?

A: That is a concept I just don?t understand: ?saving myself pain.? The only sure way to avoid all emotional pain is not to care about anything. And I can?t think of anything more painful than that.

Certainly it makes sense not to be reckless and entrust our feelings to people who?ve proven to us that they can?t be trusted ? and we should all be attuned to any self-destructive patterns in our histories.

But I don?t see either of these here. Instead I see two people who have taken two years to establish that they really like and are good for each other. Does breaking up in this case qualify as ?saving myself pain,? or ensuring it?

Meanwhile, I think you might have misunderstood my position on ?waiting.? Long-distance is sometimes unavoidable, so I don?t have a blanket opinion that waiting is a bad idea. What would paired-off service members do, for example, if their partners refused to wait out deployments?

What I do advise is to take into account: how solid the relationship is, how mutual the goals and feelings, how long you?ll be apart, and how flexible you both are about living your lives while you?re apart.

The latter has a lot of wiggle room, since some couples are committed and monogamous and should stay that way; others may be earlier in their courtship and might benefit from releasing each other from any commitment and just seeing where they are after the separation term is over. What matters is that you?re both able to think clearly instead of being all over each other about following some sort of Long-Distance Relationship Code.

When you care, you try, and you see.

LDR Veteran: My now-husband and I were in a long-distance relationship for most of two years. The advice I?d give is to not view time apart as ?waiting around? for him to get back, but instead think of it as a break from the dating world.

I felt like I got to know myself during that time even more than I did during my single years, when a lot of my mental energy was taken up by dating. And, this may sound pessimistic, but I find it really valuable to know that I can build myself a life without him if I need to.

Carolyn: Love this, thanks. I think couples also find out whether they work together on a purely conversational level ? much insight to be gained.

Texas OKs big boost in wholesale electricity rate

AUSTIN, Texas (AP) ? The Texas Public Utility Commission voted Thursday to raise the wholesale electricity price cap by 50 percent this summer in a move it hopes will spur construction of new power plants.

Texas officials say the state must boost generating capacity to meet growing demand in the next few years to help avoid rolling blackouts during peak periods such as hot summer days.

Supporters of the increase hope the lure of new profits will spur power companies to build more power plants. But consumer groups warn it could raise costs by billions of dollars, leading to higher electric bills for Texas households with no guarantee of new construction.

The vote by the three-member commission, all appointed by Republican Gov. Rick Perry, raises the wholesale rate cap on Aug. 1 from the current $3,000 per megawatt-hour to $4,500. And there could be more to come. Commissioners are already considering tripling the price to $9,000 in 2013.

The commission approved the increase on a 2-0 vote with commissioners Donna Nelson and Rolando Pablos in support. Commissioner Ken Anderson, who said he agrees with raising the cap, abstained because of concerns that this summer is too soon for some electric providers to adjust.

The cap increase affects wholesale rates within the Electric Reliability Council of Texas grid, which covers most of the state.

Texas AARP and other consumer and conservation groups say the commission hasn't studied the impact the rate increase could have on Texas households. Texas Industrial Energy Consumers, which includes refineries and chemical manufacturers, warned that a rate increase could create instability in pricing and drive smaller retail power providers out of business.

"Raising the cap this August will raise energy prices on home and business owners," said Cyrus Reed, conservation director for the Lone Chapter, Sierra Club. "It makes no sense to raise energy prices today when new generation will not result until 2014 anyway. By doing so we are just giving over our checks to electric utilities."

Pablos dismissed the warnings of skyrocketing household electric bills as "alarmist rhetoric."

"It's very clear a lot of the public don't understand the structure of the market and what we do here," Pablos said. "Energy prosperity leads to economic prosperity. We're doing this for the purpose of ensuring we have a higher quality of life for our citizens. A robust and reliable electric system will help us get there."

Although electric providers typically sign long-term contracts for electricity at a set price, they often fill in gaps on high-usage days buying wholesale on the "spot market" where prices are set several times an hour. That market can be compared to buying and selling on the stock market, where prices can be determined by sharp increases in demand and shortages.

Nelson said officials are still studying whether the change will allow electric retailers to pass the costs onto customers who already have contracts. But she said increasing the wholesale price cap by 50 percent doesn't translate into an automatic 50 percent increase on household electric bills.

Texas' competitive electricity market allows Texans to shop for the best plan they can get, she said.

"The one thing we can't do is ignore the situation and move forward blindly based on a faith and a hope we'll have enough electricity," Nelson said, pointing to the record heat that gripped the state in 2011. "If last summer taught us anything, hope for the best, plan for the worst."

Google Unveils Chrome for iPhone, iPad

Google today announced that a version of its Chrome browser for iOS will hit the App Store later today.

Chrome for iOS will work on the iPhone and iPad, Brian Rakowski, vice president of Chrome, said during the Google I/O developer conference today.

"Some of you have been very persistently asking" for Chrome on iOS, Rakowski said.

Rakowski promised a "silky smooth experience" that will allow users to close tabs with a quick swipe and swap tabs by dragging from the side. "It makes browsing the Web on your iPhone really fun," he said.

Like the desktop version of Chrome, the iOS app will sync across devices. Pull up pages that you looked at on your PC or iPhone via the iPad, or vice versa. Credentials are also synced, so no need to re-enter a password on that New York Times story via the iPad if you've already signed in on your PC.

Incognito mode is also available; toggle between public and private mode via a button on the browser bar.

The announcement comes as Google today released Chrome 20, which includes hundreds of bug fixes.
